Towelie made his first apperance on ep 805, here's breif summry of the ep:
After Cartman finds a bloody cotten thingy in Stan's bathroom, Stan's mom buys them the Okama Game Sphere.
Outside the shop, a towel comes up after they talk about something.. don't quite remember... maybe I should stop getting so high! ;) ... Anyway, Towelie askes them if they want to get high, they say no (I wounder why did they say no?) and Towelie goes away.
The boys trick their parent's so they can stay all weekend playing.
Cartman keep's making this funny comment toward's Stan's mom:
"You shouldn't have done that, he was just a'little boy!" Thinking she like aborted a baby brother to Stan or something.
Anyway, The boys are forced to go to school on Monday.
At the bus station (I think it was the bus station...) a car stops by them with a man inside. He asks them if they had seen a talking towel.
When Kyle answers; "Towelie?" The guy calls and informs someone.
After they get home from school, they find that the game sphere has been takin by the govement because they want Towelie back, and until Towelie is with them, they have their Okama game sphere.
I'm not gonna ruinn the ep by telling in detail what happens, how and why, and why during the entire ep, people keep trying to tell them what happen and no one understands that they don't care!
There are in this ep, tuns, TUNS of great lines, funny funny funny, I fell off my bunny... (WHAT??)
Stan: "Kenny, brake angrli!"
So just watch it!!

Towlie makes yet anther apperance in ep 509, where the boys go to Afganestan... Towelie makes an apperance:
Stan: "OK, thanks Towelie."
Towelie: "You wanna get high?"
Cartman: "No, we don't want to get high!!"
Towlie: "You mean, you don't want Towlie around?"
Catman: "That's right!"
Towlie: "So am I to understand that there has been a Towlie-Ban??
Hahahaha"
Boys: "OOhh!!"
Stan: "Get out of here Towelie!!!"
And there's where are little genetically engineered towel is from.
